PENGARUH EKSTRAK JINTAN HITAM (Nigella sativa) 20% TERHADAP PENURUNAN KADAR IL-6 PADA TIKUS WISTAR (Rattus norvegicus) YANG MENGALAMI PERIODONTITIS APIKALIS

Abstrak

Periodontitis apikalis adalah penyakit infeksi yang ditandai dengan responinflamasi dan destruksi tulang pada jaringan periapikal yang disebabkan olehinfeksi bakteri. Porphyromonas gingivalis, bakteri gram negatif anaerob denganaktivitas proteolitik tinggi, kandungan lipopolisakarida bakteri ini dapatmenganggu aktivasi respon pertahanan inang dengan menciptakan keradanganserta destruksi jaringan. IL-6 merupakan salah satu sitokin proinflamasi yangmemainkan peran penting dalam pathogenesis penyakit periodontal. Jintan hitam(Nigella sativa) memiliki daya antiinflamasi didapat dari kandungan tymoquinoneyang menghambat aktivasi NF-κB dan menghambat jalur siklooksigenase danlipooksigenase. Tujuan penelitian ini adalah untuk mengetahui pengaruh gelekstrak jintan hitam terhadap penurunan kadar IL-6 pada tikus wistar jantandengan periodontitis apikalis. Metode penelitian ini menggunakan eksperimentallaboratoris in vivo pada 30 ekor tikus putih galur wistar jantan dengan rancanganrandomized posttest only control group design yang diacak menjadi tigakelompok yaitu kelompok kontrol negatif: diinduksi bakteri Porphyromonasgingivalis dan diberi gel placebo, kelompok kontrol positif: diinduksi bakteriPorphyromonas gingivalis dan diberi ibuprofen, kelompok perlakuan: diinduksibakteri Porphyromonas gingivalis dan diberi gel ekstrak jintan hitam 20%. Hasilpenelitian menunjukkan terdapat perbedaan yang signifikan rata-rata IL-6 antaramasing-masing kelompok dimana didapatkan kelompok (P1) memiliki selisihterhadap kelompok (K-) yaitu sebesar -7,55 dan kelompok (K+) terhadapkelompok (K-) adalah -5,28. Dari hasil penelitian dapat disimpulkan bahwa gelekstrak jintan hitam 20% efektif dalam penurunan kadar IL-6 pada tikus wistarjantan periodontitis apikalis.

Abstract

Apical periodontitis is an infectious disease characterized by aninflammatory response and bone destruction in the periapical tissue caused bybacterial infection. Porphyromonas gingivalis, an anaerobic gram-negativebacterium with high proteolytic activity, the lipopolysaccharide content of thisbacterium can interfere with the activation of the host defense response bycreating inflammation and tissue destruction. IL-6 is one of the proinflammatorycytokines that plays an important role in the pathogenesis of periodontal disease.Black cumin (Nigella sativa) has anti-inflammatory properties obtained from thecontent of thymoquinone which inhibits the activation of NF-κB and inhibits thecyclooxygenase and lipoxygenase pathways. The purpose of this study was todetermine the effect of black cumin extract gel on reducing IL-6 levels in malewistar rats with apical periodontitis. This research method used in vivo laboratoryexperiments on 30 male white wistar rats with a randomized posttest only controlgroup design which was randomized into three groups, namely the negativecontrol group: induced by Porphyromonas gingivalis bacteria and given placebogel, the positive control group: induced by Porphyromonas gingivalis bacteria andgiven ibuprofen, the treatment group: induced by Porphyromonas gingivalisbacteria and given 20% black cumin extract gel. The results showed that there wasa significant difference in the average IL-6 between each group where group (P1)had a difference against group (K-) of -7.55 and group (K+) against group (K-)was -5.28. From the results of the study, it can be concluded that 20% black cuminextract gel is effective in reducing IL-6 levels in male wistar rats with apicalperiodontitis.
