Karies gigi merupakan salah satu masalah kesehatan gigi dan mulut yangmasih menjadi permasalahan di Indonesia hingga saat ini. Karies dapat terjadipada anak-anak maupun orang dewasa. Penyebab utama karies adalah bakteriStreptococcus mutans, Lactobacillus dan mikroorganisme lain. Pada anak-anakdengan Early Childhood Caries yang parah, Lactobacillus acidophilus ditemukansebagai penyebab dominan, mirip dengan lesi karies yang ditemukan pada orangdewasa. Lactobacillus acidophilus mempunyai peran sekunder dalam prosesterjadinya karies, karena selain bersifat asidogenik juga bersifat asidurik yangdapat hidup dalam suasana asam. Lactobacillus acidophilus hidup dalam plak dandapat merusak struktur gigi dengan mengadakan fermentasi karbohidrat. Sisikikan Barakuda mengandung alkaloid, terpenoid, saponin, flavonoid dan tanin,metabolisme sekunder tersebut pada beberapa penelitian dapat menghambatpertumbuhan bakteri Escherichia coli. Penelitian bertujuan untuk mengetahuidaya hambat sisik ikan Barakuda (Sphyraena jello) terhadap pertumbuhan bakteriLactobacillus acidophilus pada Early Childhood Caries dengan konsentrasi 50%,75%, dan 100% secara in vitro. Metode penelitian yang digunakan adalah metodedifusi untuk menentukan sensitivitas pasta sisik ikan Barakuda (Sphyraena jello)terhadap pertumbuhan bakteri dengan uji Anova One-Way. Hasil penelitiandidapatkan pasta sisik ikan Barakuda dapat menghambat pertumbuhan bakteriLactobacillus acidophilus dengan rerata zona daya hambat kontrol positif sebesar22 mm, pasta sisik ikan Barakuda 100% sebesar 17,82 mm, pasta sisik ikanBarakuda 75% sebesar 15 mm, dan pasta sisik ikan Barakuda 50% sebesar 10,4mm. Efektivitas yang paling baik dalam menghambat pertumbuhan bakteriLactobacillus acidophilus yaitu pasta sisik ikan Barakuda (Sphyraena jello)dengan konsentrasi 100%.
Abstract
Dental caries is a dental and oral health problem that is still a problem inIndonesia. Caries can occur in children and adults. The main causes of caries areStreptococcus mutans, Lactobacillus and other microorganisms. In children withsevere Early Childhood Caries, Lactobacillus acidophilus was found to be thepredominant cause, similar to the carious lesions found in adults. Lactobacillusacidophilus has a secondary role in the process of caries, because apart from beingacidogenic it is also aciduric and can live in an acidic environment. Lactobacillusacidophilus lives in plaque and can damage tooth structure by fermentingcarbohydrates. Barracuda fish scales contain alkaloids, terpenoids, saponins,flavonoids and tannins, secondary metabolism of which in several studies caninhibit the growth of Escherichia coli bacteria. The research aims to determine theinhibitory power of barracuda fish scales (Sphyraena jello) on the growth ofLactobacillus acidophilus bacteria in Early Childhood Caries at concentrations of50%, 75% and 100% in vitro. The research method used was the diffusion methodto determine the sensitivity of barracuda fish scale paste (Sphyraena jello) tobacterial growth using the One-Way Anova test. The results of the researchshowed that barracuda fish scale paste could inhibit the growth of Lactobacillusacidophilus bacteria with an average positive control zone of inhibition of 22 mm,100% barracuda fish scale paste of 17.82 mm, 75% barracuda fish scale paste of15 mm, and fish scale paste. barracuda 50% of 10.4 mm. The best effectivenessin inhibiting the growth of Lactobacillus acidophilus bacteria is Barracuda fishscale paste (Sphyraena jello) with a concentration of 100%.
