Abstract
ABSTRACT
Catharanthus roseus or tapak dara leaves are plants that have been used to treat various diseases, one of which is traumatic ulcers in the oral cavity. The healing process of traumatic ulcers requires various inflammatory mediators such as TNF-α which functions to induce epithelial growth and wound neovascularization. This study aims to examine the bioactivity of tapak dara leaf extract on the expression of Tumor Necrosis Factor-Alpha (TNF-α) in traumatic ulcer healing. This type of research is true laboratory experimental research in vivo with the post-test only control group approach. Sampling was carried out by simple random sampling technique with a total sample of 30 mice (Mus musculus). The sample consisted of five groups, namely four treatment groups and one control group which were placed in five separate cages. the research instrument used was the ELISA Kit. The average of the K(-) group was the highest compared to the other groups (289.10). ANOVA test showed that the topical bioactivity of tapak dara leaf extract gel at concentrations of 5%, 10% and 20% could reduce the expression of Tumor Necrosis Factor Alpha (TNF-α) (p value = 0.001). Based on the tukey post hoc test, the K(+) group had a better ability to reduce Tumor Necrosis Factor Alpha (TNF-α) expression in traumatic ulcer healing than the other groups (p value=0.001, mean difference=178.406). Topical tapak dara leaf extract gel can significantly reduce TNF-α expression in the healing of traumatic ulcers of the oral mucosa of mice.

Catharanthus roseus atau daun tapak dara merupakan tanaman yang telah digunakan untuk mengobati berbagai macam penyakit, salah satunya ialah ulkus traumatik pada rongga mulut, Proses penyembuhan ulkus traumatik memerlukan berbagai mediator inflamasi seperti TNF-α yang berfungsi untuk menginduksi pertumbuhan epitel dan neovaskularisasi luka. Penelitian ini bertujuan untuk menguji bioaktivitas ekstrak daun tapak dara terhadap terhadap ekspresi Tumor Necrosis Factor-Alpha (TNF-α) pada penyembuhan ulkus traumatik. Jenis penelitian ini adalah penelitian true eksperimental laboratorium secara in vivo dengan pendekatan the post-test only control group. Pengambilan sampel dilakukan dengan teknik simple random sampling dengan jumlah sampel sebanyak 30 ekor mencit (Mus musculus). Sampel terdiri dari lima kelompok, yaitu empat kelompok perlakuan dan satu kelompok kontrol yang ditempatkan di dalam lima kandang terpisah. instrument penelitian yang digunakan adalah ELISA Kit. Rerata kelompok K(-) paling tinggi disbanding kelompok lain (289,10). Uji ANOVA menunjukkan bahwa bioaktivitas topikal gel ekstrak daun tapak dara dengan konsentrasi 5%, 10% dan 20% dapat menurunkan ekspresi Tumor Necrosis Factor Alpha (TNF-α) (p value=0,001). Berdasarkan uji tukey post hoc, kelompok K(+) memiliki daya menurunkan ekspresi Tumor Necrosis Factor Alpha (TNF-α) pada penyembuhan traumatic ulcer lebih baik dibandingkan kelompok lainnya (p value=0,001, beda rerata=178,406). Topikal ekstrak gel daun tapak dara secara signifikan dapat menurunkan ekspresi TNF-α pada penyembuhan traumatic ulcer mukosa oral mencit
