Abstract
Abstrak
Karies gigi menjadi masalah kesehatan gigi dan mulut yang paling sering terjadi di Indonesia. Karies gigi merupakan penyakit multifaktor yang ditandai dengan proses demineralisasi pada jaringan keras gigi. Bakteri yang paling sering menjadi penyebab karies gigi adalah Streptococcus mutans. Pencegahan karies gigi dapat dilakukan secara kimia dengan menggunakan obat kumur, namun penggunaan yang terlalu sering tidak dianjurkan sehingga diperlukan pengembangan bahan alternatif antibakteri yang lebih aman dan efek samping yang relatif kecil. Daun sirsak (Anonna muricata L.) merupakan salah satu tanaman herbal yang dapat dikembangkan sebagai bahan alternatif antibakteri. Penelitian ini bertujuan untuk mengetahui efektivitas ekstrak daun sirsak (Anonna muricata L.) terhadap pertumbuhan bakteri Streptococcus mutans. Jenis penelitian yang digunakan adalah eksperimental laboratoris in vitro dengan rancangan penelitian Post Test Only Control Group Design. Uji daya hambat bakteri menggunakan metode difusi agar dengan cara Kirby Baurer. Penelitian ini menggunakan konsentrasi 75% dan 100% dengan kontrol positif (Chlorhexidine 0,2%) dan kontrol negatif (aquades). Berdasarkan hasil pengamatan yang dilakukan sebanyak 6 kali pengulangan, menunjukkan adanya zona bening disekitar blank disk setelah proses inkubasi selama 24 jam dengan suhu 37℃ pada inkubator. Hasil penelitian diperoleh rata – rata diameter zona hambat yang dihasilkan dari konsentrasi 100% sebesar 18,7 mm dan konsentrasi 75% sebesar 15,7 mm. Uji normalitas dengan Saphiro Wilk berdistribusi normal (p > 0,05) dan uji homogenitas dengan uji Levene bernilai tidak homogen maka selanjutnya akan dilakukan uji statistik non parametrik dengan uji Kruskal-Wallis menunjukkan perbedaan yang signifikan pada seluruh kelompok perlakuan (p < 0,05) maka dilakukan uji Post Hoc dengan uji Mann – Whitney U. Hasil uji Mann – Whitney U menunjukkan bahwa terdapat perbedaan yang signifikan pada ekstrak daun sirsak (Annona muricata L.) konsentrasi 75% dan 100% dalam menghambat pertumbuhan Streptococcus mutans. Berdasarkan hasil penelitian dapat disimpulkan bahwa ekstrak daun sirsak (Anonna muricata L.) konsentrasi 75% dan 100% efektif dalam menghambat pertumbuhan bakteri Streptococcus mutans.
Abstract
Dental caries is the most common dental and oral health problem in Indonesia. Dental caries is a multifactorial disease characterized by a demineralization process in the hard tissues of the teeth. The bacteria that most often causes dental caries is Streptococcus mutans. Prevention of dental caries can be done chemically by using mouthwash, but its use too often is not recommended so that it is necessary to develop alternative antibacterial materials that are safer and have relatively small side effects. Soursop leaf (Anonna muricata L.) is one of the herbal plants that can be developed as an alternative antibacterial material. This study aims to determine the effectiveness of soursop leaf extract (Anonna muricata L.) against the growth of Streptococcus mutans bacteria. The type of research used is experimental in vitro laboratory with Post Test Only Control Group Design. Bacterial inhibition test using agar diffusion method by Kirby Baurer method. This study used a concentration of 75% and 100% with a positive control (Chlorhexidine 0.2%) and a negative control (aquades). Based on the results of observations carried out for 6 repetitions, it showed a clear zone around the blank disk after the incubation process for 24 hours at 37℃ in the incubator. The results of the study obtained that the average diameter of the inhibition zone resulting from a concentration of 100% was 18.7 mm and a concentration of 75% was 15.7 mm. The normality test with Saphiro Wilk was normally distributed (p > 0.05) and the homogeneity test with the Levene test was not homogeneous, then non-parametric statistical tests with the KruskalWallis test would then be performed showing significant differences in all treatment groups (p < 0.05 ), then the Post Hoc test was carried out with the Mann – Whitney U test. The results of the Mann – Whitney U test showed that there were significant differences in soursop leaf extract (Annona muricata L.) at concentrations of 75% and 100% in inhibiting the growth of Streptococcus mutans. Based on the results of the study, it can be concluded that the soursop leaf extract (Anonna muricata L.) at concentrations of 75% and 100% were effective in inhibiting the growth of Streptococcus mutans bacteria.
